Flow Cytometry Core

The Flow Cytometry Core Facility offers state-of-the-art flow cytometry analysis and cell sorting for Stark Neurosciences Research Institute-affiliated researchers. Several thousand cells can be analyzed and processed using the equipment in a minute.

What can the Core do for you?

  • Help design panels and optimize experimental plans.
  • Provide consultation for data analysis.
  • Prepare and maintain the equipment for users performing FACS or flow cytometry.
  • Provides sorting and cleaning chips required for sorting.
  • Provide calibration beads for both instruments.
  • Calibrate lasers and set up the BDLSR Fortessa-X20 prior to experiments.
  • Regular courses to improve your flow cytometry.

Core Services

  • 18-parameter (16 fluorescence, FSC, SSC) cell analyzer
  • The X-20 accepts FALCON brand polystyrene 5mL FACS tubes, with an optional 96 wellplate sampler (HTS)
  • Exciter Lasers: 405, 488, 561, 638
  • Detectors
    • 405 detectors: 450/50; 525/50; 610/20; 670/30; 710/50; 780/60
    • 488 detectors: 530/30; 710/50
    • 561 detectors: 586/15; 610/20; 670/30; 710/50; 780/60
    • 638 detectors: 670/30; 730/45; 780/60

  • 14-parameter (12 fluorescence, FSC, SSC) cell sorting flow cytometer housed in a biosafety cabinet. It can also be used as a non-sorting cell analyzer.
    • 2-way sorting into 15mL conicals
    • 4-way sorting into 5mL FACS tubes
    • Well plate sorting into 6/12/24/48/96/384 well plates
  • Sorting chips are a required consumable for running the Sony, and cost $40/each. 100 micron chips (general-purpose sorting) and 130 micron chips (for large and fragile cells) are available. If you need 130 micron chips, please reach out ahead of time as these are infrequently used.  
  • The MA900 has two sets of colinear exciter lasers, pairing 405+647 and 488+561. This requires careful consideration when designing flow panels for use on the MA900.
  • Detectors
    • 488+561 detectors: 525/50; 585/30; 617/30; 695/50; 785/60
    • 405+638 detectors: 450/50; 525/50; 585/30; 617/30; 665/30; 720/60; 785/60

  • 10-parameter (8 fluorescence, FSC, SSC) cell analyzer.
  • The MACSQuant 10 is a user-friendly, highly automated instrument with the ability to self-clean and shutdown.
  • The MACSQuant will be slower, less sensitive, and less precise than the Sony MA900 or LSRFortessa X-20.
  • The MACSQuant can run96-well plates, 5mL FACS tubes, 15 and 50mL conical tubes.
  • Exciter lasers: 405, 488, 638nm
  • Detectors:
    • 405 detectors: 450/50; 525/50
    • 488 detectors: 525/50; 585/40; 655-730; 750LP
    • 635 detectors: 655-730; 750LP
    • Overnight, unsupervised runs are available at a flat fee.

  • gentleMACS Octo tissue dissociator
  • A limited set of consumables and reagents are available for new users curious about the platform. Please inquire.
  • Panel Design or Data Analysis Consultation
  • Use of Flowjo (dongle) or OMIQ data analysis software (cloud login)
